A Combination of Screening and Computational Approaches for the Identification of Novel Compounds That Decrease Mast Cell Degranulation
This study highlights the strength of integrating chemical and functional genomics screens to gain insight into the mode of action of compounds. We screened a novel small molecule library in a degranulation and in a high-content image-based endocytosis assay used for computational analysis. By correlating the endocytic profiles of the compounds with profiles from a previous screen, we identified candidate pathways including the Akt pathway and experimentally tested the predictions. Overall, we identified two novel compounds that inhibited mast cell degranulation, inhibited Akt phosphorylation, and outperformed our internal reference. Read more about this study here.
